Codon optimized Cytochrome P450 for POH synthesis
This part consists of the gene sequences for the three proteins that are needed for the synthesis of Perillyl alcohol from (-)-(S)-Limonene via the cytochrome P450 protein complex. The codon optimized sequence for Saccharomyces cerevisiae consists of the alkane hydroxylase, ferredoxin and ferredoxin reductase. The protein sequences are linked via T2A-sequences with slightly different DNA-sequences to avoid unwanted homologous recombination. The T2A-sequences allow the transcription of the three proteins from one promoter as the sequences mediate the cleavage of the peptide sequence. The cytochrome P450 was used in the project of the Marburger iGEM-Team 2016 to produce the anti-cancer drug Perillyl alcohol in Saccharomyces cerevisiae from the (-)-(S)-Limonene that has been produced in the artificial endosymbiotic E. coli.
